- Abstract: This study contains sequencing data generated to characterize CRISPR/Cas9-mediated genome editing of the myostatin (MSTN) gene in Bubalus bubalis embryos. Cas9–sgRNA ribonucleoprotein (RNP) complexes targeting exon 1 of MSTN were delivered into buffalo zygotes by electroporation. The MSTN target region was amplified from individual embryos and analyzed by Sanger sequencing to determine editing efficiency and insertion/deletion (indel) profiles. Selected embryos representing a range of Sanger-based editing efficiencies were further analyzed by targeted deep amplicon sequencing using the Illumina NovaSeq 6000 platform. The sequencing data were used to compare Sanger-based indel analysis using TIDE/ICE with next-generation sequencing-based analysis using CRISPResso2 and to characterize the diversity and frequency of CRISPR/Cas9-induced allelic variants in edited buffalo embryos.
