  • Abstract: in the present study Vibrio cholerae genome has been genetically modified to simultaneous delete four genomic islands and the TLCCTX region, without introducing any antibiotic resistance markers. These deletions rendered the strain completely avirulent. Further, heterologous expression of the Escherichia coli BL21 DE3- encoded T7 RNA polymerase in this non-toxigenic Vibrio cholerae strain BBI have conferred the strain the capacity to drive T7 promoter dependent gene expression. This enables the development of an alternative Gram-negative chassis which has a shorter doubling time. The system provides a novel controllable expression platform capable of producing higher yields of recombinant proteins.
