- Abstract: To understand the regulation of goblet cells under the regulation of NCoR1 chromatin immunoprecipitation followed by its sequencing was performed. HT29 MTX were treated with or without tricostatin A TSA for 24 hours as the treated or control groups, respectively. TSA is an universal HDAC inhibitor which inhibits the NCoR1 complex formation with HDACs and downregulates NCoR1. Briefly the cells were fixed in formalin followed by nuclear isolation. The samples were subjected to sonication. After decrosslinking, DNA was purified using PCR cleanup kit. Isolated DNA was subjected for sequencing.
