- Abstract: We performed long-read Nanopore sequencing to identify 5-methylcytosine and 5-hydroxymethylcytosine base modifications in clinical isolates of Plasmodium falciparum with diverse manifestations of malaria, including uncomplicated malaria, hepatic dysfunction, and cerebral malaria. Despite lower methylation levels globally, gene-body CpG methylation showed a consistent positive association with transcription, along with the enrichment of differentially methylated events within exons. Together, our study provides a first integrated view of DNA methylation and global transcription from clinical isolates of P. falciparum. These findings elaborate on how parasites adapt to the host environment in severe malaria and pave the way for disease-specific, informed antimalarial approaches.
