  • Abstract: Celiac disease (CD) is an autoimmune enteropathy arising in genetically susceptible individuals exposed to gluten. This study aimed to investigate the role of miRNA-mediated deregulation of the transcriptome in the seropositive and seronegative first-degree relatives (FDRs) of patients with celiac disease. We compared miRNA profiles of seropositive and seronegative FDRs of patients with Celiac Disease. Deep RNA sequencing was performed on RNA isolated from fresh duodenal biopsies. The small RNA cDNA libraries were prepared and sequenced on Illumina platform. An appropriate bioinformatic pipeline was followed. The resulting P values for differentially expressed miRNAs were adjusted for multiple testing using Benjamin and Hochberg correction for False Discovery Rate (FDR). Only miRNAs with FDR corrected P values < 0.05 and log2 fold change were considered significantly differentially expressed. Differentially expressed miRNAs in duodenal biopsies from seropositive and seronegative FDRs give an important understanding of the non-HLA miRNA-mediated pathogenesis of CD.
